Jewish settlement in Kholopenichi began in the 18th century. In the Soviet period the village had a Yiddish elementary school. Approximately half of the Jews were craftsmen. On the eve of World War II the Jewish population in Kholopenichi was 500, comprising 20.3 percent of the total population.

Kholopenichi was occupied by the Germans on July 5, 1941. All of its Jewish residents were killed in one murder operation in September 1941.

Kholopenichi was liberated by the Red Army on July 29, 1944.
